SPIO-Alginate Nanoparticles: New Platform for Calcium MR Imaging
نویسندگان
چکیده
Introduction Ca plays a vital role in many biological processes including cell differentiation and apoptosis, cardiac contractility, neurotransmission and brain function. 1 Because of the importance of Ca levels in maintaining proper function and in diseases, there is an increasing need for the development of new robust imaging platforms to perform in vivo detection of changes in Ca levels in wide range of concentrations. Currently, Ca imaging is mostly based on fluorescent methodologies. Recently, some progress has been achieved in the design of specific probes for indirect Ca MR imaging based on T1 or T2 relaxation. Combining the facts that alginates polysaccharides can be cross-linked, selectively, by Ca ions and that SPIO aggregation may change the contrast of T2* and T2 WI, we hypothesized that SPIO-alginate nanoparticles may be used as "smart" MRI probes for Ca detection (as shown schematically in Fig 1). In this study we describe the synthetic procedure of the preparation of stable water-soluble SPIO-alginate nanoparticles and the ability of these MRI probes to specifically detect different Ca concentrations in different media (water, serum and culture cell media) at physiological conditions (pH of 7.2 and 37.0C).

In vivo quantification of SPIO nanoparticles for cell labeling based on MR phase gradient images.
Along with the development of modern imaging technologies, contrast agents play increasingly important roles in both clinical applications and scientific research. Super-paramagnetic iron oxide (SPIO) nanoparticles, a negative contrast agent, have been extensively used in magnetic resonance imaging (MRI), such as in vivo labeling and tracking of cells. However, there still remain many challenge...

The prevalence of cancer has seen significant growth over the last three decades, with the American Cancer Society now reporting that one in every three Americans will be afflicted with some form of cancer in their lifetime. The debilitating effects of cancer permeate beyond obvious biological ramifications, often adversely affecting patients on emotional and financial levels as well. While sig...

Liver tumors are common and imaging methods, particularly magnetic resonance imaging (MRI), play an important role in their non-invasive diagnosis. Previous studies have shown that detection of liver tumors can be improved by injection of two different MR contrast agents. Here, we developed a new contrast agent, Gd-manganese-doped magnetism-engineered iron oxide (Gd-MnMEIO), with enhancement ef...

Purpose: Superparamagnetic iron oxide (SPIO) nanoparticles are efficient contrast agents for molecular and cellular MR imaging [1]. They are usually considered to be negative contrast agents, since their magnetic moment causes signal dephasing which results in signal voids on T2* weighted images. However SPIO agents also have intrinsic T1 shortening properties [2] that can induce signal enhance...
